Cytokine 4-Plex B
IL-17A, IL-4, IL-13, IL-5
The Simoa® Cytokine 4-Plex B (C4PB) Advantage PLUS Assay (Product # 104997) measures IL-17A, IL-4, IL-13, and IL-5 at levels as low as fg/mL, providing reliable quantification of these key proinflammatory cytokines. The levels of these 4 cytokines measured in blood offer valuable insights into inflammation and immune responses to various conditions, including infections, allergens, injuries, cancers, and immunotherapy.
The fully automated HD-X Analyzer provides unmatched scalability of up to 6 plates in an 8-hour day, accomplishing in a single day what other manual preparation methods would take a week to complete.

Key features of the Simoa® C4PB Advantage PLUS Assay
- Ultra-sensitivity: Uncover critical insights into early biological and pathological processes from low-abundant cytokines at femtogram levels.
- Multiplex: Quantify four cytokines within a single assay with 25 µL input, conserving samples and resources.
- Full Automation: Enhance efficiency and reproducibility with walk-away automation on the HD-X.
- Scalability: Adapt the automated workflow for fast time-to-results or high-throughput, generating data from one 96-well plate in under 3 hours or up to 6 plates daily.
- Lot-to-Lot Consistency: Improve reliability for longitudinal and large-scale studies.
The target cytokines of Simoa® C4PB Advantage PLUS
IL-17A: IL-17A is produced by T helper type 17 (Th17) cells and drives proinflammatory responses by recruiting monocytes and neutrophils to inflammation sites. It is associated with various immune and autoimmune diseases, including rheumatoid arthritis, asthma, lupus, allograft rejection, and psoriasis.
IL-4: IL-4 is expressed by T helper type 2 (Th2) cells, mast cells, basophils, and eosinophils. It promotes cell proliferation, survival, class switching to IgG4 and IgE in B cells, and facilitates Th2 differentiation. IL-4 plays a central role in allergic inflammation and asthma by activating mast cells, eosinophils, basophils, and epithelial cells.
IL-13: IL-13 is secreted mainly by activated T-helper type 2 cells. It is a key mediator in allergic asthma, regulating eosinophilic inflammation, mucus production, and airway hyperresponsiveness. IL-13 also influences apoptosis and tumor growth in cancers such as B-cell chronic lymphocytic leukemia and Hodgkin’s disease.
IL-5: IL-5 is produced by T-cells and is critical for B-cell differentiation into antibody-secreting cells. It also plays a key role in eosinophil biology, driving eosinophil differentiation, maturation, and migration. By promoting eosinophil proliferation and activation, IL-5 is a major contributor to allergic reactions and asthma symptoms.
Applications and Intended Use
